We haven't done this for a while. Some of my favorites:
N28 Anson. No resemblance to the player in the N300 or the N162. T206 Johnson pitching. T206 Mathewson portrait. I don't see it. I've seen the photo it probably is based on and it just doesn't capture it. CJ Jackson. Weak resemblance at best; compare to the photo cards. 33 Goudey Ott especially green background. The red one isn't that great a likeness either, to my eye. 34 Gehrig 61. Compared to the 37, a weak likeness. 38 Goudey Greenberg. I could be wrong, but i don't see it. 52T Mantle. Seriously. Put aside the preconceptions and LOOK at it.

In agreement about that N28 Anson and the 34 goudey gehrig #61. N28 is a great set with great looking lithographs, but that Anson card is horrid compared to the rest in that set.

Any M.P. & Co. probably takes the cake, though probably does not fit in the OP's stated criterion of being major cards.

Here is a classic one. The ironic thing is that Irv (pictured here) actually does resemble Cy a little bit. If this player were right handed you could almost not know it's not Cy....

I believe lot #1 in the current B&L auction would fit this thread. ... I would also say (most) strip cards, but I'm guessing they're not considered "major."

Yes! All the E-91s! Which in actuality depicts the Campbell's soup kid in various poses rather than the players.

He was probably referring to the fact that Joe, in a real picture, looks like he chewed on rocks his whole life and was hit by a train...but yeah, you could say it's a different shade of lipgloss.

There's a Frank Chance card, I can't remember the series, that shows him from behind with his head turned. I'm a die hard Cubs fan but I don't want a card of Frank Chance's ass
